Abstract—An irreversible chemical transformation in an enzyme–substrate complex is accompanied by iso-
tope fractionation that only simulates thermodynamic equilibrium but is determined by the kinetic isotope
effect. The conclusion is drawn that elementary chemical transformations in enzyme–substrate complexes are
irreversible.
